Description
Vascular endothelial growth factor (VEGF) is a key regulator of angiogenesis and vasculogenesis. It belongs to a family of secreted growth factors that stimulate the proliferation, migration, and survival of endothelial cells.
VEGF exerts its biological activity through binding to VEGF receptor tyrosine kinases (VEGFR-1/Flt-1 and VEGFR-2/KDR) located on endothelial cells. Activation of these receptors triggers intracellular signaling pathways including MAPK, PI3K/AKT, and PLCγ, leading to endothelial cell proliferation, vascular permeability, and formation of new blood vessels.
VEGF is essential for vascular development, wound healing, and tissue regeneration, and dysregulated VEGF signaling is implicated in tumor angiogenesis and inflammatory diseases.
Multiple isoforms of VEGF exist due to alternative splicing, including VEGF121, VEGF165, and VEGF189 in humans. The canine VEGF120 isoform represents the shorter soluble form lacking heparin-binding domains, resulting in increased diffusibility in the extracellular environment.

After refolding, the recombinant protein is predominantly present in the dimeric form, although SDS-PAGE analysis may reveal both monomeric (~15 kDa) and dimeric (~30 kDa) species even under reducing conditions.

Biological Activity
The biological activity of recombinant VEGF is determined using a cell proliferation assay with endothelial cells, typically human umbilical vein endothelial cells (HUVEC). This endothelial cell proliferation assay is widely used as a standard functional test for VEGF bioactivity, reflecting activation of VEGF receptor signaling pathways.
