Description
Prolactin (PRL) is a pituitary peptide hormone of the somatotropin/prolactin family that plays a central role in the regulation of mammary gland development and lactation. In addition to its classical lactogenic function, prolactin is involved in reproduction, growth, development, and osmoregulatory and immunomodulatory processes.
Prolactin exerts its biological effects through binding to the prolactin receptor (PRLR), a class I cytokine receptor, leading to activation of intracellular signaling pathways that include JAK2/STAT5 and that regulate proliferation, differentiation, and hormone-responsive gene expression.

Biological Activity
The biological activity of recombinant bovine prolactin is determined using a cell proliferation assay with Nb2-11 rat lymphoma cells. In this assay, prolactin stimulates proliferation of Nb2-11 cells in a dose-dependent manner, and activity is quantified by measuring cell growth relative to untreated controls. Nb2/Nb2-11 cells are a well-established bioassay system for lactogenic hormones such as prolactin, and this assay is widely used in the literature and by cell repositories for prolactin bioactivity testing.
Specific activity / ED50: To avoid overstatement, this should be determined experimentally for the final recombinant bovine construct and lot. Commercial recombinant prolactin products from other species often report Nb2-11-based ED50 values, but those values should not be assigned directly to this bovine construct without in-house testing.
