Description

Brain-derived neurotrophic factor (BDNF) is a member of the neurotrophin family of growth factors, which also includes nerve growth factor (NGF), neurotrophin-3 (NT-3), and neurotrophin-4/5 (NT-4/5). BDNF plays an essential role in the survival, differentiation, and maintenance of neuronal populations in both the central and peripheral nervous systems. It is widely expressed in the brain, particularly in the hippocampus, cortex, and cerebellum.

BDNF promotes neuronal survival, neurite outgrowth, synaptic plasticity, and neuronal differentiation. Its biological effects are mediated primarily through activation of the TrkB receptor tyrosine kinase, which initiates intracellular signaling pathways involved in neuronal growth and survival.Alterations in BDNF expression or signaling have been associated with several neurological disorders, including neurodegenerative diseases and mood disorders.

Quality Control

Biological Activity

The biological activity of recombinant human BDNF can be determined using cell-based assays that measure TrkB receptor activation.

Common bioassays include:

  • Cell proliferation assays using BaF3 cells engineered to express TrkB

  • Neuronal survival assays

  • Neurite outgrowth assays in primary neuronal cultures

These assays evaluate the ability of BDNF to activate TrkB-mediated signaling pathways that promote neuronal survival and differentiation.
